Regulation of Gadd45γ expression by C/EBP

摘要: The Gadd45γ (growth arrest and DNA damage-inducible) gene is activated transcriptionally by at least two kinds of agents: damaging agent such as methyl methanesulfonate (MMS) UV radiation, or cytokines interleukin (IL)-6, IL-2 granulocyte colony-stimulating factor (G-CSF). To investigate the sequences transcription factors involved in induction after treatment with IL-6, human was cloned sequenced. We found C/EBP (CCAAT/enhancer-binding protein) family proteins, major IL-6 signal transduction pathway, could regulate transcriptional activity promoter. In addition, a noncanonical C/EBP-binding site within promoter where C/EBPβ C/EBPδ bind, identified electrophoretic mobility shift assay (EMSA) reporter analysis. Furthermore, we coordinated expression profile between mRNA C/EBPs (β δ) protein during differentiation M1 cells: amount transcripts became maximal when both levels were high, on day 1 cells IL-6. These findings suggest that mitotic growth coupled to cell mediated stimulation arrest-associated genes Gadd45γ.
