摘要: Abstract Specific binding of triamcinolone acetonide was analyzed in cytosols from developing mouse visceral yolk sac and fetal, neonatal adult liver. In the sac, capacity increased 1 × 10 3 sites/cell on day to maximal levels (9 sites/cell) 14 gestation. fetal liver, sites were low (2 18, rapidly after birth approx. 1.7 4 by 9 postpartum present at Scatchard analysis data indicated presence a single class cytosolic limited high affinity ( K d = 2–4 nM). The level specific nuclear 2 h injection [ H]triamcinolone proportional number for each tissue tested. physicochemical characteristics glucocorticoid-receptor complexes examined DEAE-Sephadex A-50 column chromatography. “Unactivated” liver eluted 0.4 M KCl. Heat “activated” 0.25 KCl, whereas those prewash fractions (0.02 potassium phosphate buffer). These results provide evidence that quantitative but not qualitative changes glucocorticoid receptors occur during development also establish midgestation sac.
