摘要: Isolated hepatocytes incubated in the presence of NO donors S-nitroso-N-acetylpenicillamine (SNAP) and 3-morpholino-sydnonimine (SIN-1) displayed a time- dose-dependent inhibition glucose synthesis from lactate plus pyruvate as substrate which correlated with production, but not nitrite production. Neither parent compound SNAP, N-acetyl-DL-penicillamine (NAP), nor or nitrate had any significant effect on output, indicating that was due to generation within incubation medium. The concentrations required for this (< 800 nM) are range reported occur intact tissues vivo. magnitude inhibitory SNAP (approximately 50%) comparable endotoxin treatment rat substrate. When number different substrates examined, showed pattern observed after rat, suggesting may be mediator effects bacterial hepatic gluconeogenesis. donor no flux through 6-phosphofructo-1-kinase, supporting concept primary site gluconeogenesis by both resides at level phosphoenolpyruvate formation.
