摘要: Background: Ischemic stroke is the second leading cause of death and disability worldwide, which needs to develop new pharmaceuticals for its prevention treatment. Qingda granule (QDG), a traditional Chinese medicine formulation, could improve angiotensin II-induced brain injury decrease systemic inflammation. In this study, we aimed evaluate neuroprotective effect QDG against ischemia/reperfusion-induced cerebral illustrate potential mechanisms. Methods: The middle artery occlusion/reperfusion (MCAO/R) surgery in vivo oxygen-glucose deprivation/reoxygenation (OGD/R) vitro models were established. infarct volume was quantified using magnetic resonance imaging (MRI). Neurobehavioral deficits assessed five-point scale. Cerebral histopathology determined by hematoxylin-eosin (HE) staining. Neuronal apoptosis evaluated TUNEL immunostaining with NeuN antibodies. protective on OGD/R-injured HT22 cells MTT assay Hoechst 33258 expression lncRNA GAS5, miR-137 apoptosis-related proteins investigated MCAO/R-injured rats RT-qPCR western blot analysis. Results: significantly reduced ischemic volume, accompanied improvements neurobehavioral deficits. Additionally, ameliorated histopathological changes neuron loss rats. Moreover, improved growth inhibited injured OGD/R vitro. Finally, decreased Bax cleaved caspase3, whereas it increased Bcl-2 cells. Conclusion: plays role via regulation GAS5/miR-137 signaling pathway.
