A comparative study of disease genes and drug targets in the human protein interactome

摘要: Disease genes cause or contribute genetically to the development of most complex diseases. Drugs are major approaches treat disease through interacting with their targets. Thus, drug targets critical for treatment efficacy. However, interrelationship between and is not clear. In this study, we comprehensively compared network properties five categories (cancer, cardiovascular disease, immune system metabolic nervous disease). We first collected from genome-wide association studies (GWAS) corresponding drugs based on drugs' Anatomical Therapeutic Chemical (ATC) classification. Then, obtained these different categories. found that, though intersections were small, significantly enriched in enrichment human protein-coding genes. further proteins encoded by protein-protein interaction networks (interactome). The results showed that tended have higher degree, betweenness, lower clustering coefficient cancer Furthermore, observed a clear fraction increase near neighborhood randomized study presents comprehensive comparison context interactome. provide some foundational characteristics designing computational strategies predict novel repurposing.
