摘要: Application of alginate-polylysine (PLL) capsules for immunoisolation living cells are suffering from a varying degree success and large lab-to-lab variations. In this study we show that these differences in rates can be attributed to alginate dependent essential physicochemical changes the properties in vivo will render more susceptible inflammatory responses. Capsule were studied before after implantation by XPS, immunocytochemistry, measuring zeta potentials. We capsule type which provokes unknown reasons strong response, i.e. high-guluronic (G) with near identical but evokes minimal intermediate-G capsules. The cause difference response was decrease nitrogen content on high-G due detachment PLL an increase zeta-potential. Our data illustrate important overlooked phenomena; not necessarily exposure microenvironment might induce undesired responses failure encapsulated cellular grafts.
