摘要: Abstract Racemization of amino acids was studied in alkali treated β-casein, oat protein (Avena sativa) and rapeseed (Brassica napus). Amino acid hydrolyzates containing enantiomeric acids, L-Xxx D-Xxx, were converted into mixtures N-ethoxycarbonyl-(Eoc-) derivatized diastereomeric dipeptide pairs, Eoc-Phe-L-Xxx, Eoc-Phe-D-Xxx Eoc-Val-L-Xxx, Eoc-Val-D-Xxx upon the reaction with ethoxycarbonylphenylalanine N-hydroxysuccinimide ester (Eoc-Phe-ONSU) ethoxycarbonyline (Eoc-Val-ONSU), respectively. The epimeric products separated determined by reversed phase high-performance liquid chromatography content D-isomers calculated. β-casein using this method agreement gas chromatography-mass spectrometry techniques described literature. In proteins exposed to alkaline conditions for 3 24 hours 0.1 N NaOH at 65°C, extend racemization followed sequence Ser>Asp>Phe>Glu>Val most samples. preliminary experiments, alkali-treated sunflower seed (Helianthus annuus) samples examined D-Phe. reported HPLC technique represents a simple practical assessment dietary
