摘要: Objective To investigate the DNA methylation using pyrosequencing and its effects on upregulation of IL6 mRNA in patients with recurrent aphthous stomatitis (RAS) connection hematinic deficiency atopy. Material Methods. This cross-sectional study was conducted at Dr. Hasan Sadikin Hospital, Bandung, from January-March 2019 approved by Health Research Ethics Committee Universitas Padjadjaran (Ethics No. 990/UN6.KEP/EC/2018). Furthermore, subjects had RAS ulcers a history least twice year along atopy dietary imbalance no intraoral herpes or any systemic diseases. performed 23 21 healthy subjects, sampling carried out consecutively. The blood samples were collected all then, RNA extracted peripheral mononuclear cells (PBMCs). Consequently, bisulfite-modified used to confirm status gene promoter through method. levels assessed reverse transcriptase-polymerase chain reaction technique. expression control group analyzed 2-ΔΔCT statistical analysis Mann-Whitney U test evaluate p value <0.05 considered be statistically significant. Result approximately 1.88-fold patients, there significant relationship between increased risk (p < 0.001). It reported that four six sites cytosine phosphate guanine (CpG) island lower degree methylation, two other greater compared control, but not Conclusion showed control. present is 566-658, whereas location 1-1684. Thus, it would necessary conducting some research CpG islands determine methylation.
