Investigating retinal toxicity of a lutein-based dye in a model of isolated and perfused bovine retina.

摘要: Retidyne™ is a new lutein-based dye for internal limiting membrane staining. It uses the intrinsic staining characteristics of lutein which already known to act as an antioxidant and blue-light filter in human retina. We investigated retinal tolerance different times measured by electroretinogram (ERG) isolated perfused retina whole mount. For functionality, testing bovine retinas were prepared with oxygen saturated standard solution ERG was recorded until stable b-wave amplitudes reached. Then perfusion stopped applied directly onto surface exposure 60 or 120 s. After restarting solution, monitored 75 min. To investigate effects on photoreceptor function alone, 1 mM asparate added block b-waves. time 60 s a- b-waves remained throughout experiment. Exposure 120 s caused initial drop that reached statistical significance only a-waves (a, − 21%, p = 0.047; b, − 14%, p = 0.052). This effect seen during first minutes washout recovered completely. In model retina, showed good safety profile common intraoperatively used times. An toxic regarding transient cannot be ruled out but might also explained partial blockage flashlight due more intense at beginning washout.
