摘要: This work was targeted at characterizing the effect of high pressure on kinetics lysozyme crystallization via a series experiments and mathematical modeling. Pressure effects nucleation crystal growth were distinguished by evaluating rate constants for two kinetic processes various pressures. In first phase work, examined with primary objective increasing crystallization. Lysozyme carried out initially concentration 40 mg/mL, pH 4.5 0.8 M sodium chloride pressures ofO.l, 50, 100 150 MPa. A factorial experiment performed level time pressurization as factors four levels each factor. Protein recovery in crystals atmospheric significantly different from all pressure-treated samples. An overall enhancement could be obtained period followed pressure. Initial duration elevated either 10 min or 2 h total 36 (including pressurization) resulted rates that 2-3 times higher than those without pressurization. not evident lower protein concentrations 19 28 mg/mL. Enzymatic activity unchanged solubility appeared result similar behavior observed closer to iso-electric point (pH 9.5), i.e. gave
