摘要: Abstract Introduction Resveratrol, a naturally-occurring phytophenol, has been shown to bolster immune surveillance and reverse immunosenescence in dose dependent manner rodents humans. Although safety pharmacokinetic studies have completed dogs, the immunomodulatory effects of resveratrol dogs not yet investigated. The objective this study was determine effect on canine innate system function vitro. hypothesis that similar other species, low concentrations would stimulate while high depress function. Methods Whole blood collected from six healthy, adult, client-owned incubated with at final 6000 ng ml−1, 3000 ng ml−1, 1000 ng ml−1, or control solution for 4 h. Following incubation, phagocytosis oxidative burst were evaluated using flow cytometry, LPS-, lipoteichoic acid (LTA) – peptidoglycan (PG)-stimulated leukocyte production TNF, IL-6, IL-10 measured specific multiplex assay. Results Phagocytosis altered by any concentration compared control. However, number PMNs capable performing did change, robustness reaction following stimulation Escherichia coli PMA reduced manner. In addition, LTA-, PG, PBS-stimulated TNF increased incubation all control, dependent. LTA-stimulated IL-6 Furthermore, decreased 6000 ng ml−1 3000 ng ml−1 PG-stimulated TNF:IL-10 ratio lower concentrations. Conclusion While sparing PMN phagocytosis, it burst. Resveratrol also pro-inflammatory anti-inflammatory cytokine capacity These data suggest supplementation may reactions promoting decreasing production. Based these findings, further vivo regarding capability are indicated prior routine supplementation.
