The behavior of Bupleurum scorzonerifolium as a parent of somatic hybrid introgressed lines is associated with UV resistance of its chromosomes

摘要: Protoplasts of Bupleurum scorzonerifolium irradiated with 380 μW/cm2 UV for 5 min were fused by the PEG-mediated method untreated protoplasts Arabidopsis thaliana. The fusion products cultured in P5 liquid medium single hybrid cell clone formation. As a total, 81 independent putative clones (cell lines) obtained, and seventeen them identified as somatic hybrids chromosome counting, GISH, RAPD, SSR analyses. More than 80 B. scorzonerifolium-like green introgressed plants leaves regenerated from 49 lines, which contained chromatin DNA characteristic A. To assess tolerance both parents exclusion introgression, their UV-irradiated (380 0 min), thaliana more sensitive to those judged Single Cell Gel Electrophoresis analysis. possible relationship between resistance elimination formation is discussed.
