Identification of pemphigus vulgaris antigen extracted from normal human epidermis and comparison with pemphigus foliaceus antigen.

摘要: Immunoprecipitations of cultured keratinocyte extracts have shown that pemphigus vulgaris (PV) sera bind a polypeptide 210,000 mol wt with disulfide-linked chains 130,000 and 85,000 wt. To identify proteins in normal human skin recognized by PV antibodies, we performed immunoprecipitations epidermal extracts. All 22 tested immunoprecipitated complex polypeptides (PV complex) 210,000, 130,000, wt, after reduction. One- two-dimensional gel electrophoresis showed the 130,000- 85,000-mol-wt antigen from both keratinocytes epidermis identical charges sizes. In addition to precipitating complex, 14 also antibodies calcium-sensitive epitope on different (PF which has previously been be precipitated all foliaceus (PF) sera. The PF consists 260,000, 160,000, 110,000, Although majority precipitate no complex. Thus, can absolutely distinguished molecular level using patients' autoantibodies. complexes, although distinct, certain similarities. each is identical. 160,000-mol wt-peptide 130,000-mol-wt peptide same isoelectric point are capable disulfide linkage polypeptide. complexes closely related may prove important cell adhesion.
