Down-regulation of FUT3 and FUT5 by shRNA alters Lewis antigens expression and reduces the adhesion capacities of gastric cancer cells

摘要: Abstract Background Lewis antigens are fucosylated glycoconjugates involved in the development of several pathologies. The adhesion sialyl-Lewis to E-selectin is a key step metastasis and glycosidic component CD44 plays role binding hyaluronic acid, extracellular matrix associated tumor invasion. Fucosyltransferases enzymes that add fucose precursor glycan structures: FUT3 FUT5 catalyze addition α1-3,4 position detected epithelial cells. In this study, we have analyzed effects silencing FUT3, or FUT3/FUT5, two gastric cancer cell lines, expression adhesive migratory capacities Methods FUT3/FUT5 were down-regulated using lentiviral delivery shRNAs MKN45 GP220 Results infected cells, decreased levels mRNA by quantitative RT-PCR; lower antigens, evaluated flow cytometry, observed. endothelial cells trough E-selectin, acid reduced shFUT3, shFUT5 shFUT3/FUT5, whereas CD44, western blot, did not change. General significance down-regulation reduces cells; allows identify specific fucosyltransferases implicated synthesis cellular model.
