The Y-Box Binding Protein 1 Suppresses Alzheimer’s Disease Progression in Two Animal Models

摘要: The Y-box binding protein 1 (YB-1) is a member of the family DNA- and RNA proteins. It involved in wide variety DNA/RNA-dependent events including cell proliferation differentiation, stress response, malignant transformation. Previously, YB-1 was detected neurons neocortex hippocampus, but its precise role brain remains undefined. Here we show that subchronic intranasal injections recombinant YB-1, as well fragment YB-11−219, suppress impairment spatial memory olfactory bulbectomized (OBX) mice with Alzheimer’s type degeneration improve learning transgenic 5XFAD used model cerebral amyloidosis. YB-1-treated OBX showed decreased level β-amyloid. In animals, an improved morphological state revealed hippocampus; mice, delay amyloid plaque progression observed. Intranasally administered penetrated into could enter neurons. vitro co-incubation monomeric β-amyloid (1–42) inhibited formation fibrils, confirmed by electron microscopy. This suggests interaction prevents filaments are responsible for neurotoxicity neuronal death. Our data first evidence potential therapeutic benefit treatment disease.
