Understanding electron transport systems of Streptomyces cytochrome P450

摘要: Streptomyces spp. are known to produce various types of biologically active compounds including antibiotics, antiparasitic agents, herbicides and immunosuppressants. P450 (cytochrome P450) enzymes may have key roles in these biosynthetic biotransformation reactions. Recent genomic analysis coelicolor A3(2) indicates that S. six ferredoxins (Fdxs), four putative Fdx reductases (FdRs) 18 genes. However, there few clues explain the mechanisms functions systems. To solve questions, we expressed purified five P450s, FdRs Fdxs Escherichia coli. Of CYP105D5 has fatty acid hydroxylation activity a system reconstituted with putidaredoxin reductase Fdx4 or spinach FdR Fdx, although reconstitutions FdR2 FdR3 any did not support CYP105D5-catalysed oleic hydroxylation. Elucidation detailed electron transport for provide perspective usefulness P450s as biocatalyst.
