摘要: The presence of amylosucrase in 12 Alteromonas and Pseudoalteromonas strains was examined. Two species (Alteromonas addita KCTC 12195 macleodii 2957) possessed genes that had high sequence homology to known amylosucrases. Genomic clones containing the ASase analogs were obtained from A. macleodii, deduced amino acid sequences corresponding (aaas amas, respectively) revealed they highly similar ASases Neisseria polysaccharea, Deinococcus radiodurans, geothermalis. Functional expression amas Escherichia coli successful, typical activity detected purified recombinant AMAS, whereas AAAS nonfunctional. Although maximum total AMAS observed at 45 degrees C, ratio transglycosylation increased as temperature decreased 55 25 C. These results imply occurs preferentially lower temperatures while hydrolysis is predominant higher temperatures.
