Molecular cloning of ESET, a novel histone H3-specific methyltransferase that interacts with ERG transcription factor.

摘要: The ets-related gene erg encodes a transcription factor that is implicated in the control of cell growth and differentiation. To identify interacting partners ERG, we screened yeast two-hybrid cDNA library constructed from mouse hematopoietic cells using N-terminal region ERG as bait. We isolated 4.6 kb full-length encoding 1307-amino acid protein migrating 180 kD band, which was termed ESET (ERG-associated with SET domain). 92% identical to human SETDB1 (SET domain, bifurcated 1). interaction between supported by vitro pull-down glutathione S-transferase (GST) fusion protein, transfection co-immunoprecipitation experiments, association endogenous ERG. Since possesses evolutionarily conserved SET, preSET, postSET domains histone methylation, tested ability methylate core histones. results these studies demonstrated H3-specific methyltransferase, mutations within abolished its methyltransferase activity. Together, findings raise possibility may participate transcriptional regulation through ESET-mediated methylation.
