Novel signal amplification approach for HRP-based colorimetric genosensors using DNA binding protein tags.

摘要: The need for sensitive detection of DNA is growing as more specific sequences are being correlated to gene markers disease diagnosis, food safety and other security related applications. Detection in hybridization-based assays usually achieved with target-specific ssDNA probes conjugated directly enzyme labels like HRP that provide signal amplification or nanoparticles functionalized multiple molecules. In order overcome some the drawbacks presented by these approaches, we developed a unique sensing platform based on an HRP-DNA binding protein tag conjugate hybrid ssDNA-dsDNA probe. Specifically, this work describe preparation characterization scCro its application model target sequence. By using HRP-scCro together probe containing three scCro-specific dsDNA sites, demonstrate improvement over 3-fold both sensitivity limit high-risk human papillomavirus (HPV16), compared standard ssDNA-HRP conjugate. These results show can be used alternative universal tool enhancement enzyme-linked suitable integration point-of-care systems.
