摘要: 1. This study was designed to determine whether clofilium exhibits antifibrillatory activity in a pinacidil + hypoxia-induced model of ventricular fibrillation (VF) Langendorff-perfused hearts. 2. Ten minutes after exposure vehicle or (0.1, 1.0 and 10.0 microM), hearts were exposed (1.25 then subjected 12 min hypoxia reoxygenated. Onset VF recorded. Additional groups pretreated with UK-68,798 (1.0, 3.0 delayed rectifier channel blocker, 5-hydroxydecanoate (10 known ATP-dependent K+ an identical protocol. 3. Clofilium decreased the incidence concentration-dependent manner; 7/9 control developed vs 1/9 (P = 0.007, Fisher's Exact) treated microM clofilium. In addition, protected from VF, while pretreatment did not. 4. separate group hearts, electrically-induced converted sinus rhythm 10/11 introduced as bolus. 5. is capable preventing rabbit isolated heart manner. We have data suggest that ability attenuate effects pinacidil+hypoxia our may include blockade metabolically active channels, i.e., KATP (glibenclamide-sensitive) channel.
