摘要: Transgenic tobacco plants containing maize gene (Z4) encoding a 23-kd zein protein, which is normally synthesized in the endosperm of seeds, were obtained using modified Ti plasmid vector. Although polyadenylated transcript homologous to Z4 was present seeds some these transgenic plants, protein could not be detected any tested (35 total). To simplify analysis tissue specificity promoter (Z4(pro)) different organs transformed additional chimeric genes Z4(pro)-CAT and Z4(pro)-GUS produced. Very weak seed-specific CAT activity observed one out ten Z4(pro)-CAT-transformed plants. When more sensitive GUS assay system used evaluate Z4(pro) tobacco, it shown all 11 that restricted seeds. study synthesis stability proteins protein-coding region also cDNA clone (A30) 19-kd placed under control 35S (35S(pro)) cauliflower mosaic virus. Undegraded both size classes roots, leaves tissue, but embryos, mature from 35S(pro)-zein-transformed The did appear broken down during seed germination; zeins began green cotyledons.
