Transcriptomic analysis in the developing zebrafish embryo after compound exposure: individual gene expression and pathway regulation.

摘要: The zebrafish embryotoxicity test is a promising alternative assay for developmental toxicity. Classically, morphological assessment of the embryos applied to evaluate effects compound exposure. However, by applying differential gene expression analysis sensitivity and predictability may be increased. For defining signatures toxicity, we explored possibility using exposures based on commonly expressed individual genes as well regulated pathways. Four toxic compounds were tested in concentration-response design, caffeine, carbamazepine, retinoic acid valproic acid, two non-embryotoxic compounds, d-mannitol saccharin, included. With transcriptomic analyses able identify genes, which mostly development related, after exposure embryotoxicants. We also identified pathways embryotoxicants, suggestive their modes action. Furthermore, whereas all within these can differ each compound. Overall, present study suggests that use pathway regulation useful starting points biomarkers predicting embryotoxicity.
