Growth inhibition of human endothelial cells by human recombinant tumor necrosis factor alpha and interferon-gamma.

摘要: We investigated the effect of recombinant tumor necrosis factor-alpha (rTNF-alpha) on proliferative response human umbilical vein endothelial cells (HUVEC) to normal serum (NHS), in absence or presence interferon (IFN)-gamma. rTNF-alpha significantly impaired NHS-stimulated HUVEC growth at a dose as low 0.1 U/ml. The inhibitory was dose-dependent up 50-100 U/ml and already evident after 2 h incubation. Doses range 10 completely suppressed 3H-thymidine uptake stimulated by 7.5% NHS, partially overcome 10-20% NHS. not cytotoxic doses 1000 rIFN-gamma also effective suppressing incorporation, (0.1 U/ml) rTFN-alpha showed an additive effect. TFN-alpha IFN-gamma antagonizing vascular endothelium variety factors contained could be relevant pathologic conditions involving damage proliferation.
