摘要: Chromosome 1 is involved in quantitative anomalies 50-60% of breast tumours. However, the structure these and identity affected genes remain to be determined. To characterise define their consequences on gene expression, we undertook a study combining array-CGH analysis expression profiling using specialised arrays. Array-CGH data showed that 1p was predominantly losses 1q almost exclusively gains. Noticeably, high magnitude amplification infrequent. In an attempt fine map regions copy number changes, defined 19 shortest overlap (SROs) for gains (one at 18 1q) 20 SROs (all 1p). These SROs, whose sizes ranged from 170 kb 3.2 Mb, represented smallest genomic intervals possible based resolution our array. The elevated incidence 1q, added well-established concordance between DNA increase augmented RNA made us focus changes this chromosomal arm. identify candidate oncogenes, studied profiles 307 located home-made built cDNA We identified 30 showing significant overexpression correlated increase. order substantiate involvement, levels were measured by (Q)-RT-PCR panel 25 cancer cell lines previously typed array-CGH. Q-PCR 11 significantly overexpressed presence gain lines, when compared normal breast.
