Comparison of minidogfiler and "ASCH" STR multiplex systems for preliminary estimation of variability within wolf´s like dog breeds

摘要: Abstract This study was undertaken to estimate basic population parameters and compare resolution capabilities of two chosen STR panels (Mini-DogFiler, MDF; “ASCH”). Wolf´s like dog breeds, namely Czechoslovakian wolfdog (n = 55), Saarloos (n = 22), samples the Eurasian wolf (n = 21) were selected for their close genetic relationship. Also, German Shepherd (n = 26) breed included as a donor genome wolfdog. Overall 228 alleles detected using 20 markers (MDF 11 “ASCH” 9 loci). Number ranged from 3 23 per locus, with median locus MDF 10 “ASCH”. Both showed similar level variability in means number (k) observed heterozygostity (HO) there not found statistically significant differences (Mann-Whithey U-test; p = 0.85 k, p = 0.88 HO). A group wolves highest unlike very low within all measured parameters, although such only k parameter panel (Kruskal-Wallis ANOVA; p = 0.0003) no other groups different. microsatellite able correctly distinguish breeds wolfs verified by STRUCTURE programme several potential hybrids identified. Moreover, this pilot analysis amenable creation allelic ladders thus ensure consistency reliability future extended samples.
