摘要: The agent responsible for prion diseases is a misfolded form of normal protein (PrPC). hypothesis stipulates that PrPC must be present the disease to manifest. Cervid populations across world are infected with chronic wasting disease, horizontally-transmissible likely spread via oral exposure infectious prions (PrPCWD). Though PrPCWD has been identified in many tissues, there little effort characterize overall expression cervids and its relationship accumulation. We used immunohistochemistry (IHC), western blot enzyme-linked immunosorbent assay describe naive white-tailed deer. real-time, quaking-induced conversion (RT-QuIC) detect seeding activity CWD-infected assessed tissues comprising alimentary tract, alimentary-associated lymphoid tissue systemic from 5 was expressed all though often very low compared level CNS. IHC specific cell types wherein high. To compare distribution PrPCWD, we examined deer advanced CWD infection. Using RT-QuIC, detected 21 tissues. In 3 subclinical sacrificed 4 months post-inoculation, consistently tissue, irregularly tract not at brain. Contrary our levels dictate accumulation, higher lower gastrointestinal than system salivary glands oropharyngeal tissue. These data suggest sole driver accumulation accumulate before centrifugal brain occurs.
