摘要: AIM: To investigate the effects of transforming growth factor β1 (TGF-β1) on differentiation colonic lamina propria fibroblasts (CLPF) into myofibroblasts in vitro. METHODS: Primary CLPF cultures were incubated with TGF-β1 and analyzed for production α-smooth muscle actin (α-SMA), fibronectin (FN) FN isoforms. Migration assays performed a modified 48-well Boyden chamber. Levels total phosphorylated focal adhesion kinase (FAK) after induction migration. RESULTS: Incubation 2 d did not change α-SMA levels, while treatment 6 significantly increased production. Short term incubation (6 h) enhanced migration, long d) reduced migration to 15%-37% compared untreated cells. isoform mRNA expression short (2 contrast d. After TGF-β1-preincubated showed higher amounts its isoforms lower levels FAK than cells. CONCLUSION: Long induced α-SMA, migratory potential phosphorylation, In contrast, contact dose-dependent increase cell phosphorylation without
