摘要: Long double-stranded RNAs (dsRNAs) are duplex that can induce immune response when present in mammalian cells. These historically associated with viral replication, but recent evidence suggests human cells naturally encode endogenous dsRNAs regulate antiviral machineries cellular contexts beyond response. In this study, we use photochromic organic compound spiropyran to profile and quantitate dsRNA expression. We show the open form of spiropyran, merocyanine, intercalate between RNA base pairs, which leads protonation alteration spectral property compound. By quantifying change, detect quantify expression level, both synthetic cellular. further demonstrate spiropyrans be used as a molecular diagnostic tool endogenously expressed dsRNAs. Particularly, robustly elevated levels colorectal cancer treated 5-aza-2′-deoxycytidine, an FDA-approved DNA-demethylating agent for chemotherapy, thus demonstrating predicting responsiveness drug treatment. As signature virus accumulation is implicated various degenerative disease, our work establishes potential application simple diagnose disease based on
