In vitro DNA binding studies of antiretroviral drug nelfinavir using ethidium bromide as fluorescence probe

摘要: Understanding the interaction of small molecules with DNA has become an active research area at interface between biology and chemistry. In present work, we investigated mode nelfinavir (NFV) herring sperm (hs DNA) under physiological conditions using various biophysical techniques. Analysis UV-absorption fluorescence spectra indicates formation complex NFV hs DNA. According to results, binding constant (K) was found be 3.30 × 10(4)LM(-1). The calculated thermodynamic parameters (ΔH° ΔS°) suggested that hydrogen bonding plays a major role in them. Phosphate group studies revealed there no electrostatic interactions occurred Circular dichroism (CD) melting curve were employed measure conformational change presence NFV, which verified minor groove mode.
