摘要: Bronchial tissue kallikrein is the major kininogenase activity in airways of asthmatic subjects. The relationship IgE-mediated events to its release and/or activation unknown, however, and subject this report. Seven subjects with mild atopic asthma underwent endobronchial challenge relevant aeroallergen. Baseline pre-allergen lavage sequential post-challenge lavages were collected over an approximate 10-minute time course. Individual aliquots analyzed separately compared saline control performed a separate lobe. In five seven subjects, increase activity, measured by cleavage synthetic substrate Val-Leu-Arg-pNA, was identified lavages. antigenic identity enzymatic confirmed as each case immunoblotting. Tissue highly correlated appearance immunoreactive histamine kinin (p = 0.0001). High molecular weight kininogen influx detected samples immunoblotting paralleled detection BAL fluid. Two despite clinical profiles similar those positive responders, failed react challenge. Saline contained detectable kallikrein, kinin, two subjects; case, significantly less than post-allergen samples. results demonstrate close association between immediate type hypersensitivity lower airway active substrate, liberation kinin.
