Generation and Characterization of B72.3 Second Generation Monoclonal Antibodies Reactive with the Tumor-associated Glycoprotein 72 Antigen

摘要: Abstract Monoclonal antibody (MAb) B72.3 was generated using a membraneenriched fraction of human mammary carcinoma biopsy. It has demonstrated reactivity to the majority adenocarcinomas including colorectal, gastric, pancreatic, ovarian, endometrial, mammary, and non-small cell lung cancer no or weak normal adult tissues, with exception secretory endometrium. The B72.3-reactive antigen, termed tumor-associated glycoprotein (TAG)-72, been purified used as an immunogen generate second generation MAbs. Since source TAG-72 colon (CC) xenograft, these MAbs have given CC designation. Twenty-eight MAbs, all immunoglobulin Gs, shown be reactive via both radioimmunoassay immunohistochemical analyses show differential versus tissue biopsies. Nine (CC11, 15, 29, 30, 40, 46, 49, 83, 92) were selected for further characterization. As result direct-binding range carcinomas, Western blotting, live surface binding assays, five liquid competition radioimmunoassays, K measurements, nine could distinguished from each other B72.3. determined 2.54 × 10 9 m -1 ; higher s CC92, 83 having 14.26, 16.18, 27.72 , respectively. These studies thus demonstrate that one more anti-TAG-72 may efficient than B72.3, useful in combination toward study population diagnostic therapeutic procedures presently utilizing MAb
