Ultrasensitive electrochemical detection of Pb2+ based on DNAzyme coupling with exonuclease III –Assisted target recycling

摘要: Abstract A sensitive and selective electrochemical Pb2+ sensor was developed based on GR-5 DNAzyme exonuclease III-assisted target recycling amplification. When existed, cleaved into two free DNA fragments. One of the fragments could open hairpin (HP1) modified electrode to form duplex with blunt end at 3′-terminus triggered Exo III cleavage process. large amount residual part HP1 were obtained by III-aided which hybridized another labeled methylene blue, resulting in dramatic increase current. The signal blue positive correlated concentration. proposed has concentration ranging from 10−11 8×10−6 mol/L a detection limit 2.74 p mol/L. Moreover, analysis real water revealed that biosensor promising application environment monitoring.
