摘要: Chemoresistance is a poor prognostic factor in breast cancer and, thus, presents significant clinical challenge. The mechanisms of chemoresistance involve multiple complex biological processes. This study aims to identify common contributory factors by comparing protein expression profiles chemosensitive MCF-7 cells and resistant two different commonly used anti-cancer drugs (adriamycin paclitaxel). Expression the ATP binding cassette transporter, P-glycoprotein (P-gp), tumours has previously been found correlate with prognosis vivo accordingly, we confirmed overexpression P-gp both adriamycin- paclitaxel-resistant cells. Using two-dimensional gel electrophoresis MALDI-TOF peptide mass fingerprinting, identified 20 proteins differentially expressed between chemosensitive, adriamycin-resistant Cytokeratin-8, keratin-19, Hsp-27, 14-3-3 epsilon, annexin-A2 phosphoglycerate kinase-1 showed altered Validation number these changes was Western blotting. Our findings provide further insights into chemoresistance, as well representing an attractive starting point for identification potential biomarkers predict response chemotherapy vivo.
