Lysozyme Enhances the Inhibitory Effects of the Peroxidase System on Glucose Metabolism of Streptococcus mutans

摘要: The combined effect of the salivary peroxidase system and lysozyme on glucose uptake Streptococcus mutans NCTC 10449 was investigated. bacteria were grown to late-exponential phase, washed, re-suspended in buffer at pH6, incubated with (1) 50 micrograms/mL from human milk for 60 min; (2) 7-15 mumol/L hypothiocyanous acid/hypothiocyanite 10 (3) min prior addition incubation min. Glucose initiated by adding bacterial suspensions mL pre-warmed containing 0.98 D-(U-14C-)-glucose, mixture a shaking water-bath 37 degrees C. Samples withdrawn various time intervals, rapidly filtered through 0.45-microns membranes, washed ice-chilled buffer, incorporated radioactivity determined. Lysozyme stimulated S. slightly, but significantly inhibited rattus metabolism. A 20-30% inhibition radiolabeled incorporation observed alone. Incubation resulted total uptake. In contrast, combination without gave only 30-50% inhibition. 5 mmol/L dithiothreitol after eliminated viability not affected treatment any components used. Our results indicate that physiological concentrations have synergistic which significant metabolism mutans.
