Direct regeneration from in vitro leaf and petiole tissues of Populus tremula ‘Erecta’

摘要: Dormant buds from a mature tree of Populus tremula ‘Erecta’ were incubated on Murashige and Skoog (MS) medium supplemented with 1.0 μM thidiazuron (TDZ). Induced shoots then proliferated MS or Woody Plant Medium (WPM), Driver Kuniyuki Walnut (DKW) varying levels benzyladenine (BA). Overall, grown 1.25–2.5 BA exhibited the highest frequency shoot proliferation (>95%) more than 60% responding explants produced five per explant. Shoot organogenesis was induced both leaf petiole WPM containing BA, TDZ, zeatin. Among different cytokinins tested, zeatin (average 72.1%) organogenesis. None survived media no within 6–8 weeks following culture. higher regeneration obtained petioles explants. The achieved when 10–20 Addition naphthaleneacetic acid (NAA) did not have significant effect in all treatments. directly without intervening callus. Regenerated easily rooted tested 0.5 NAA. Rooted plants transferred to potting mix greenhouse.
