摘要: This report describes a sensitive, simple and rapid direct-binding solid phase sandwich radioimmunoassay for measurement of intrinsically labeled immunoglobulins synthesized in vitro . With the direct technique, small volumes tissue culture supernatants or cell sonicates can be placed directly assay tubes and, after short incubation washing, bound specific antigen counted. new complements previously described inhibition immunoglobulin quantitation. use both techniques on same samples, detailed quantitative characterization cellular synthesis, assembly secretion obtained under variety circumstances. The quantity, absolute activity antigenic composition determined routinely with application two techniques. A series experiments plasma cells that synthesize monoclonal IgG illustrate applications methods. are applicable to detection unlabeled nanogram microgram range suited studies synthesis where such sensitivity specificity required.
