摘要: The function of ARHGEF10, a known guanine nucleotide exchange factor (GEF) for RhoA with proposed roles in various diseases, is poorly understood. To understand the precise this protein, we raised monoclonal antibody against ARHGEF10 and determined its localization HeLa cells. was found to localize vesicles containing Rab6 (of which there are three isoforms, Rab6a, Rab6b Rab6c), Rab8 two Rab8a Rab8b), and/or secretion marker neuropeptide Y (NPY)-Venus Rab6-dependent manner. These were originate from Golgi contain secreted or membrane proteins. Ectopic expression an N-terminal-truncated mutant led generation large vesicle-like structures both Rab8. Additionally, small interfering (si)RNA-mediated knockdown impaired these exocytotic vesicles. Furthermore, invasiveness MDA-MB231 cells markedly decreased by as well From results, propose that acts exocytosis tumor invasion Rab8-dependent
