摘要: The direct myocardial action of daunomycin and related compounds was studied in artificial perfusate or blood-perfused isolated dog hearts. Samples for paper thin-layer chromatography were collected at intervals 1, 5, 15, 30 60 minutes. In these experiments (50 mg) caused an increase coronary perfusion pressure the same time a metabolite, daunomycinone, formed. When hearts perfused with Ringer9s solution minutes dose only slight increases little daunomycinone injection 50 18 mg into heart preparations immediate pressure. Adriamycin produced effects on vasculature almost identical to daunomycin, whereas N-acetyldaunomycin no deleterious effects. found be broken down daunomycinone-like product manner similar significantly less conversion detected. These tend indicate that significant amount adriamycin is converted blood this metabolite may responsible artery constriction observed preparations.
