摘要: Abstract Introduction: Oligonucleotide microarray analysis can be powerful tool to determine gene expression changes after transplantation. However, the genes analyzed by this technique in transplanted organs do not provide information regarding specific cell types. Moreover, since are infiltrated with recipient lymphocytes, accurately reflect either donor or infiltrating lymphocytes. To obviate difficulty we used recently developed of laser microdissection (LMD) obtain endothelial cells from heart allografts undergoing ischemia/ reperfusion injury (I/R). Methods: Endothelial were dissected cryostat sections coronary vessels C57 BL/6 (H-2b) donors and Balb/C (H-2d) recipients using Leica® LS AMD dissector. RNA was subjected three rounds aRNA amplification on an Affymetrix MG-U74A mouse Gene Chip®. Genes showing stastically significant control normal mice further for functional pathways effects Simplified Ontology Kyoto Encyclopedia Genomes public databases. Results: LMD yielded 550–600 20 hearts. Adequate amount obtained analysis. Ninety-eight upregulated 172 downregulated (p Conclusion: A combination functionally based is a method analyze types
