摘要: 1. Head injury is the major cause of death and severe disability in young adults. Evidence from clinical studies shows ischaemic brain damage to be determinant bad outcome, that a proportion this (perhaps up 40%) delayed, thus offering an opportunity for 'prophylactic' therapy. 2. Laboratory several relevant animal models human head (fluid percussion, subdural haematoma, focal ischaemia by middle cerebral occlusion) indicate excitatory amino acids are important mediators damage. Pretreatment with NMDA antagonists has shown extent may dramatically reduced these (68% reduction cat MCA occlusion model, 54% rat haematoma model). 3. Trials therefore strongly indicated.
