Knockdown of sal-like 4 expression by small interfering RNA induces apoptosis in breast cancer cells.

摘要: Breast cancer is the most prevalent cancers worldwide and causes a significant amount of deaths annually. Spalt-like transcription factor 4 known as factor, which has an important role in proliferation cancerous cells. Small interfering RNA (siRNA) short-chain molecule 20 to 25 nucleotides that protrude on two sides 3', nucleotides. In this study, using specific sequence siRNA against gene, its activity investigated cell line breast cancer. The cells (MCF-7) were cultured then, anti-sal-like (SALL4) siRNA, their toxic doses determined. Then, gene transfected into cell. Proliferation expression SALL4 BCL-2 measured real-time polymerase chain reaction method. concentration IC50 was 40.35 nmole. Gene results indicated Bcl-2 group significantly reduced ( P < 0.05). SiRNA can increase apoptosis by reducing Bcl-2; it be used novel targeted therapy. This strategy, addition increasing specificity drug, also reduces side effects when compared with conventional chemotherapy.
