Caveolin-1 mediates chemoresistance in cisplatin‑resistant ovarian cancer cells by targeting apoptosis through the Notch-1/Akt/NF-κB pathway

摘要: Caveolin-1 (Cav-1), a family of ubiquitously expressed oligomeric structural proteins in many mammalian cells, has been shown to be an effective regulator tumorigenesis. Recent studies have indicated that Cav-1 can promote resistance chemotherapy variety tumors. However, the regulation on chemoresistance ovarian cancer is still unknown. In present study, mRNA and protein expression level was investigated by RT-PCR western blot analysis, respectively, 50% inhibitory concentration (IC50) value measured MTT assay. The P-glycoprotein (P-gp), Notch-1, p-Akt p-NF-κB p65 were detected using analysis apoptotic ratio determined Annexin V-FITC/PI detection kit. results showed levels significantly higher SKOV3/DDP A2780/DDP than SKOV3 A2780, respectively. Knockdown decreased IC50 cisplatin-resistant cells. P-gp significant had no correlation with siRNA transfection. induced cisplatin normal cells knockdown could enhance cell apoptosis. Furthermore, also able downregulate Overexpression upregulated value, but under effect Notch-1 or LY294002 PDTC, markedly decreased. Our suggested targeting apoptosis through Notch-1/Akt/NF-κB pathway.
