DNA polymorphism of leptin gene in Bos indicus and Bos taurus cattle

摘要: Leptin plays an important role in the regulation of feed intake, energy metabolism, growth and reproduction cattle. We used polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique to screen for DNA polymorphisms leptin gene 403 cattle belonging various breeds Bos indicus (Hariana, Sahiwal, Gir Nimari cattle), taurus (Holstein Friesian (HF) Jersey cattle) as well x crossbreds (½ HF ½ Hariana). In all we amplified two regions gene, a 522 bp comprising partial intron 2 exon 3 another 94 consisting part 2. Digestion PCR products with BsaAI restriction enzyme revealed three genotypes studied. This is first report presence purebred Indian origin (indicine cattle). Almost similar genotype frequencies were observed breed groups, while frequency mutant homozygotes (AA) was very low (0.03 0.07). On digestion Kpn2I enzyme, HF, crossbred The CC had highest (0.68) whereas CT heterozygotes (0.69) mutation absent indicine results suggest that BsaAI-RFLP has occurred far back evolution before divergence taurine recently indicated by fact this only detected
