Ultrasound microbubbles mediated miR-let-7b delivery into CD133+ ovarian cancer stem cells.

摘要: Ovarian cancer stem cells (OCSCs) are considered the reason for ovarian cancer’s emergence and recurrence. Ultrasound-targetted microbubble destruction (UTMD), a non-vial, safe, promising delivery method miRNA, is reported to transfect (CSCs). In present study, we investigated miR-let-7b into OCSCs using UTMD. The CD133+ OCSCs, accounted only 0.1% of cell line A2780, were separated by flow cytometry, CSC characteristics have been proved spheroid formation self-renewal assay. transfection efficiency UTMD was significantly higher than other groups except lipofectamine group through cytometry. viability all decreased after transfection, late apoptosis rate miR-let7b induced 2.62%, while that non-treated 0.02% (P<0.05). Furthermore, Western blot results demonstrated surface marker CD133 expression has decreased. Therefore, our indicated UTMD-mediated miRNA could be platform therapy.
