摘要: OBJECTIVES A modafinil daily dosing strategy promotes wakefulness in narcolepsy patients experiencing excessive daytime sleepiness; however, some may continue to experience late-day sleepiness. Excessive sleepiness is associated with cognitive impairment. Modafinil has improved executive function other models of This study evaluated the effects once-daily vs. split doses on and combined despite satisfactory treatment earlier day. METHODS After a 2-week washout, 24 received 3 weeks double-blind 400-mg once (7 AM) plus placebo (noon) or 600-mg dose (400 mg, 7 AM; 200 noon). Assessments included Maintenance Wakefulness Test (MWT) for individual regimens Wisconsin Card Sort (WCST) treatments combined. RESULTS was significantly more effective than improving MWT scores (5 PM-7 PM; P < 0.05). Significant mean (+/- SEM) reductions from baseline 8.2 +/- 2.7 total number errors 5.9 1.9 percent (P 0.05, both) were demonstrated WCST. well tolerated; adverse events headache (n = 1), emotional lability bronchitis accidental injury 2), no reports insomnia. CONCLUSIONS For residual narcolepsy, an additional 200-mg taken at midday sustaining throughout entire waking Treatment also function.
