摘要: We have developed an easy-to-operate and effective method for performing the sequential online analysis of enzyme reactions based on capillary electrophoresis (CE). The system was constructed by passing two capillaries through a sample vial at distance 5 μm between ends. Direct injection CE were achieved periodically switching high-voltage power supply off on, without any physical disturbance inlet. injected via concentration diffusion with in-column derivatization amino acids occurring interface capillaries. High reproducibility injections demonstrated relative standard deviation values (n = 20) 1.01%, 1.25%, 0.80% peak height, area, migration time, respectively. Sequential assay glutamate pyruvate transaminase catalyzed reaction carried out simultaneously monitoring substrate consumption product formation every 30 s from beginning to end reaction. Michaelis constants obtained found be in good agreement results traditional off-line assays. Our has great potential usage chemical analytes ultraviolet or laser-induced fluorescence detection.
