摘要: Abstract A partial cDNA clone for the potato wound-inducible metallocarboxypeptidase inhibitor (PCI) was isolated from a library constructed mRNA of abscisic acid (ABA)-treated leaves. The full 5′ region obtained through RACE-PCR protocol. PCI encodes precursor polypeptide which comprises 29 residue N-terminal signal peptide, 27 pro-region, 39 mature protein, and 7 C-terminal extension. Northern blot analysis demonstrates that gene is transcriptionally activated by wounding, wound signaling can be induced ABA jasmonic acid. Subcellular localization protein investigated immunocytochemistry electron microscopy, showing accumulates within vacuole. form, comprising extension, has been expressed in Escherichia coli characterized. Its inability to inhibit carboxypeptidases, stability carboxypeptidase digestion, suggest pro-domain may have, besides probable vacuolar sorting function, role modulation inhibitory activity PCI.
