Quantitative metabolic network profiling of Escherichia coli: An overview of analytical methods for measurement of intracellular metabolites

摘要: Understanding the mechanism of metabolic network response to extracellular changes has become one primary objectives systems biology. Quantitative profiling model organisms such as E. coli is principal approach. Well-designed analytical protocols for determination intracellular metabolites may provide a crucial premise quantitative cellular metabolism. This article reviews and evaluates existing methods determining in terms sample preparation mass spectrometry (MS) analysis. Critical issues including variation status interference external pools during cell sampling, loss quenching procedures, degradation efficiency extraction, matrix effects separation quantification are discussed. Moreover, suggestions provided solve problems based on results from recent studies order establish precise reliable platform metabolites.
