摘要: Background: Measurement of estrogen receptor (ER), progesteron (PR) and human epidermal growth factor 2 (HER2) status in early breast cancer is critical for informing treatment recommendations. Targetprint®, a commercially available microarray-based test, measures mRNA levels ER, PR HER2 genes. The aim this study was to investigate the concordance accuracy local clinical subtyping by inmunohistochemistry (IHC) fluorescence situ hybridization (FISH) with TargetPrint®. Material Methods: We collected data retrospectively from 109 patients 17/5/2012 17/4/2015. All them underwent surgery. were assessed IHC tumor samples. For 2+ cases, additional FISH used. These results compared microarray quantifications. Microarrays all performed formalin-fixed paraffin-embedded (FFPE) Accuracy TargetPrint® evaluated positive (PPV) negative (NPV) predictive value considering as "Gold Standard". Corcondance between techniques percentage Cohen9s κ coefficient. interpretation k Coefficient done correlating its qualitative scale (Landis Koch, 1977): 0 considered poor; 0,01-0.20 slight; 0,21-0,40 fair; 0,41-0,60 moderate; 0,61-0,80 substantial; 0,81-1 almost perfect. Results: 100% samples RE both resulted negative, 3 positive. Regarding RP, 80% positive, 11% 5% 4% 100%, k=1. PR, 90,83%, k=0,65 (95%CI 0,45-0,85). In case HER2, 97,25%, k=0 (kappa paradox). TargetPrint had PPV 1 NPV assesing ER. 0,93 0,75. 0,97. Conclusions: To best our knowledge, first exploring IHC/FISH Targetprint® FFPE According previous fresh tissue, perfect ER well substantial seen. suggest lack technique, test or intra-tumor heterogeneity possible reasons less consistent accordance PR. It would be interesting further design prospective address question.
