摘要: Abstract The scientific and medical history of valproic acid is relatively long, compared with other frequently used psychopharmacologic agents. Valproic was as an organic solvent in research laboratories for eight decades, until the fortuitous observation action against pentylenetetrazol-induced convulsions rodents. Early clinical experience emphasized therapy absence seizures primary generalized epilepsies. During two decades controlled trials partial-onset generalized-onset myoclonus, valproate established prototypical broad-spectrum antiepileptic drug. Anecdotal observations patients both epilepsy migraine headaches who were started on led to prospective, randomized that antimigraine efficacy. suggested antimanic actions; more than a decade later, significant efficacy mania. Antiproliferative effects unexpectedly noted during mechanistic studies; later maintenance adjunctive or chemopreventive role oncology being defined. While pharmacokinetic studies appear definitive, completion comprehensive pharmacodynamic investigations valproate's biochemical actions utility yet be achieved.
